Background and Additional Info

Background: The VHL-BRD4 Proximity/Competition Assay Kit enables identification of compounds that bind the canonical VHL ligand-binding site by disrupting a degrader-induced ternary complex. This assay uses the VHL-based degrader MZ1 to induce proximity between recombinant VHL / ELOB / ELOC and BRD4, which brings Alpha-compatible donor and acceptor beads into close proximity and generates a measurable signal. Test compounds that bind VHL competitively displace MZ1, reducing the proximity signal and allowing rapid identification of VHL ligands. The kit includes optimized recombinant proteins, assay buffers, detection beads, and reference controls to support robust proximity-based screening in a 384-well format. Signal changes corresponding to disruption of the VHL-BRD4 complex are quantified using an Alpha-compatible plate reader.
