CRBN ligands were compared following the instructions provided in the TAK-107 kit. Assay performance included a S:B ratio of 88:1 and Z' = 0.91.

Background and Additional Info

Background: The CRBN-BRD4 Proximity/Competition Assay Kit enables rapid identification of compounds that bind the canonical cereblon (CRBN) ligand-binding site through disruption of a degrader-induced ternary complex. The assay uses the degrader dBET1 to induce proximity between recombinant CRBN / DDB1 and BRD4, bringing Alpha-compatible donor and acceptor beads into close proximity and generating a measurable signal. Test compounds that bind CRBN competitively displace dBET1, reducing the proximity signal and enabling straightforward identification of CRBN ligands. The kit includes optimized recombinant proteins, assay buffers, detection beads, and reference controls for robust proximity-based screening in a 384-well format. Results are quantified using an Alpha-compatible plate reader by monitoring signal changes corresponding to disruption of the CRBN-BRD4 complex.
