Recombinant Human Tetra-Ubiquitin Chain Proteins Set

Protein Sets

Product Code: TPS-161
Verified Applications: Ubiquitin chains exhibit diversity in length, linkage type, and associated cellular functions. M1-, K6-, K11-, K29-, K33-, K48-, & K63-linked Tetra-Ubiquitin serve as valuable reagents in assays involving ubiquitin-binding proteins and as a substrate for ubiquitin-specific deubiquitylating enzymes (DUBs).
Activity: Polyubiquitin chains can serve as substrates in in vitro reactions with deubiquitylating enzymes (DUBs) that hydrolyze bonds between adjacent ubiquitin molecules. Furthermore, polyubiquitin chains can be utilized to explore the mechanisms of binding and recognition between these chains and other proteins with UIMs, UBAs, and/or other ubiquitin-binding motifs.

Background and Additional Info

Background: The Tetra-Ubiquitin Chain Proteins Set includes seven recombinant, linkage-defined tetra-ubiquitin (tetra-Ub) chains representing M1, K6, K11, K29, K33, K48, and K63 ubiquitin linkages. Each chain consists of four ubiquitin monomers connected through a defined linkage type, enabling the investigation of chain length- and linkage-dependent recognition within the ubiquitin–proteasome system (UPS). These distinct polyubiquitin architectures participate in diverse cellular processes, including proteasomal degradation, inflammatory signaling, DNA damage responses, and protein quality control. Compared with shorter ubiquitin chains, tetra-ubiquitin provides an extended polyubiquitin architecture for studying multivalent interactions with ubiquitin-binding domains, ubiquitin receptors, and deubiquitylating enzymes (DUBs). This set supports comparative characterization of linkage-selective binding, enzymatic chain processing, and polyubiquitin recognition mechanisms, making it valuable for biochemical assay development and drug discovery. Each linkage is supplied as an individual 10 µg aliquot, providing a standardized collection for investigating polyubiquitin signaling across seven biologically relevant linkage types. Set contents: 7 × 10 µg aliquots | Total protein: 70 µg
