Background: SMARCA2 bromodomain (BD) is the catalytic ATPase subunit of the SWI/SNF (BAF) chromatin remodeling complex, where it regulates chromatin accessibility and transcription through ATP-dependent nucleosome repositioning. Isoform 2 of the bromodomain recognizes acetylated lysine residues on histone tails, directing recruitment of the remodeling complex to chromatin and contributing to the regulation of gene expression. SMARCA2 bromodomain has emerged as an important target for epigenetic therapeutics, including bromodomain inhibitors and targeted protein degraders that exploit synthetic lethal interactions in SMARCA4-deficient cancers.
