Recombinant Human Di-Ubiquitin Chain Proteins Set

Protein Sets

Product Code: TPS-160
Verified Applications: Ubiquitin chains exhibit diversity in length, linkage type, and associated cellular functions. K6-, K11-, K29-, K33-, K48-, & K63-linked Di-Ubiquitin serve as valuable reagents in assays involving ubiquitin-binding proteins and as a substrate for ubiquitin-specific deubiquitylating enzymes (DUBs).
Activity: Polyubiquitin chains can serve as substrates in in vitro reactions with deubiquitylating enzymes (DUBs) that hydrolyze bonds between adjacent ubiquitin molecules. Furthermore, polyubiquitin chains can be utilized to explore the mechanisms of binding and recognition between these chains and other proteins with UIMs, UBAs, and/or other ubiquitin-binding motifs.

Background and Additional Info

Background: The Di-Ubiquitin Chain Proteins Set includes six recombinant, linkage-defined di-ubiquitin (di-Ub) chains representing K6, K11, K29, K33, K48, and K63 ubiquitin linkages. Each chain consists of two ubiquitin monomers connected through a specific lysine-mediated isopeptide bond, providing a defined reagent for investigating linkage-dependent signaling within the ubiquitin–proteasome system (UPS). Different ubiquitin chain linkages regulate distinct cellular processes, including proteasomal degradation, DNA damage responses, intracellular trafficking, and protein homeostasis. This set enables comparative analysis of ubiquitin-binding proteins, deubiquitylating enzymes (DUBs), and other components of the ubiquitin signaling machinery across six linkage types. The defined chain architecture makes these reagents particularly useful for evaluating linkage selectivity, characterizing protein–ubiquitin interactions, and developing biochemical and biophysical assays. Each linkage is supplied as an individual 10 µg aliquot, providing a convenient, ready-to-use collection for ubiquitin signaling research and drug discovery applications. Set contents: 6 × 10 µg aliquots | Total protein: 60 µg
