BpJOS exhibits broad clippase activity toward diverse ubiquitin chain linkages. K6-, K11-, K29-, K33-, K48-, and K63-linked diubiquitin (Ub₂) and M1-linked tetraubiquitin (Ub₄) were incubated with BpJOS for 1, 10, and 30 min and analyzed by SDS–PAGE. BpJOS cleaved all tested Ub₂ linkages, producing monoubiquitin (Ub), while M1-linked Ub₄ was progressively converted to lower molecular weight ubiquitin species.
