Background: SMARCA4 bromodomain (BRG1 BD) is a key acetyl-lysine recognition module within the catalytic ATPase subunit of the SWI/SNF (BAF) chromatin remodeling complex, where it contributes to ATP-dependent regulation of chromatin accessibility and transcription. By binding acetylated lysine residues on histone tails and other chromatin-associated proteins, the bromodomain helps recruit and position the remodeling complex at specific genomic loci to control gene expression. SMARCA4 bromodomain has emerged as an important target for epigenetic drug discovery, including the development of bromodomain inhibitors, molecular glues, and targeted protein degraders aimed at modulating SWI/SNF complex function in cancer and other diseases.
