Background: The Ubiquitin K-Only Mutant Proteins Set contains a set of mutant ubiquitin proteins in which only one canonical lysine residue is preserved (K6, K11, K27, K29, K33, K48, or K63), while all other lysines are mutated to arginine. These “lysine (K)-only” mutants restrict ubiquitin chain elongation to a single defined linkage site, enabling precise characterization of ubiquitin chain topology. By limiting polyubiquitin assembly to one lysine position at a time, this kit allows for the determination of the linkage specificity of E2 conjugating enzymes and E3 ubiquitin ligases. These reagents are particularly valuable for identifying the preferred ubiquitin linkage types formed during enzymatic reactions and for validating mechanistic models of ubiquitin signaling. The kit supports systematic interrogation of ubiquitin chain architecture and provides a controlled framework for studying linkage-dependent signaling outcomes. These proteins are well suited for in vitro ubiquitylation assays, enzyme characterization, and studies of ubiquitin linkage biology.
