Background and Additional Info

Background: DCAF11 (DDB1- and CUL4-associated factor 11) is an adaptor protein for the CUL4-DDB1 E3 ubiquitin ligase complex, mediating substrate recognition and ubiquitination. It contains WD40 repeats enabling protein–protein interactions and targets diverse substrates for proteasomal degradation, regulating processes such as cell cycle progression, DNA damage response, and chromatin remodeling. DCAF11 influences replication stress responses and transcriptional control by recruiting specific factors to CUL4 complexes. Dysregulation or altered expression of DCAF11 has been linked to impaired genome stability and may contribute to disease states, including cancer. Ongoing studies aim to delineate substrate networks and therapeutic potential of modulating DCAF11 activity.
