Background: UBE2R1 (also known as CDC34-like E2 enzyme, UBC3/UBE2R1) is an E2 ubiquitin-conjugating enzyme that mediates transfer of activated ubiquitin to substrate proteins in cooperation with E3 ligases. It plays a central role in the ubiquitin–proteasome system, regulating cell-cycle progression, signal transduction, and protein quality control by promoting polyubiquitination and subsequent proteasomal degradation. UBE2R1 activity influences degradation of cyclins and CDK inhibitors, impacting G1/S transition and genomic stability. Dysregulation of UBE2R1 has been implicated in cancer and neurodegenerative disorders. Its enzymatic mechanism and interactions with specific E3s make it a candidate for targeted modulation in disease contexts.
