Background and Additional Info

Background: UCHL1 (PGP9.5) is a neuron-enriched cysteine deubiquitylase that recycles ubiquitin by cleaving small adducts fused to the c-terminal glycine 76 of ubiquitin, sustaining proteasomal proteostasis in axons and synapses. It is highly expressed in central/peripheral neurons and some neuroendocrine tissues and testes. UCHL1 supports axonal transport, synaptic function, and neuronal survival; rare mutations (e.g., I93M) and dysregulation are linked to Parkinson’s disease and other neurodegenerative conditions. It is also overexpressed in several cancers and serves as a tissue and fluid biomarker of neuronal injury.
