Background and Additional Info

Background: DDB1- and CUL4-associated factor 16 (DCAF16) is the substrate recognition subunit of the CRL4-DCAF16 cullin E3 ubiquitin ligase complex. It facilitates the ubiquitylation and subsequent proteasomal degradation of the nuclear protein Spindlin-4 (SPIN4), which may be the only natural substrate for this ligase. Structurally, DCAF16 consists of a compact α-helical bundle and a short C-terminal acidic tail that mediates DDB1 binding. DCAF16 residues Cys177, Cys178 and Cys179 are frequent interaction sites for electrophilic ligands used to target FKBP12, PARP2, CDK4/6, BRD4, and BRD9 for ubiquitylation and subsequent proteasomal degradation, demonstrating broad scope and potential for DCAF16 in the field of Targeted Protein Degradation (TPD).
